酮洛芬凝胶的制备及体外透皮试验
)置25ml容量瓶中,用0.01mol/LNaOH溶液稀释至刻度,于260nm波长处测定吸收度值:以空白对照品同法操作所得结果进行空白校正。校正后的吸收度值代人标准曲线方程计算样品浓度,并计算累积渗透量O(含每次取样时的损失量)。5.5结果(见表1)结果表明含薄荷脑的酮洛芬凝胶具有良好的透皮效果(P<0.01)。6讨论酮洛芬为一疏水性物质,因而不易溶于水TheJournalofamacemi∞】PracticeVo1.171999No.5性凝胶。本文利用其酸性.采用乙醇溶解,并用三乙醇胺中和至中性的方法,增加了酮洛芬在凝胶中的稳定性。经过冷热试验及留样观察.未见酮洛芬析出,含量稳定。但酮洛芬见光易氧化变色,故本制剂应密闭避光保存。表1酮洛芬凝肢体外透皮累积渗透量Q(mg,±5,n=5)处方中薄荷脑为促渗剂,体外透皮试验显示薄荷脑具有较强的促渗作用,但因薄荷脑不溶于水,直接加人凝胶基质中会导致其分布不均,故宜采用乙醇溶解后在不断搅拌下加入的方法。
